No, you shouldn't get cranky. That's actually pretty evolved sibling loyalty going on there. Depending on the three-year-old's understanding and ability to have a discussion, you might want to talk about better ways of handling conflict than hitting (enlist help of adult). But I'd praise that child to the skies for caring about her sister and intervening, even if her way of intervening was a little kid's physical way.
